Flammulina velutipes

Flammulina velutipes is an edible basidiomycete also called golden needle mushroom with increasing agricultural and biotechnological importance, However, genetic manipulation and breeding of this organism are restricted because of the lack of knowledge about its genomic information.
The aim of this project is whole genome sequencin of Korean F.velutipes and func-tional genomics based on its sequernce infomtoin.
An electrophoretic karyotype of Korean F.velutipes was obtained using contour-clamped homogeneous electric field(CHEF) gel electrophoresis.

The monokaryotic progenies(4019-20 and 4019-19) and a dikaryotic strain (4019-2018) had 6-8 chromosomes, with sizes ranging from 1.6 to 5.8Mb, Among the 3 strains that were examined, strain 4019-20 resolved into at least 7 chromosomes, with a total genome size of approximately 26.7MB. In order to determine the whole genome sequence, we constructed a bacterial artificial chromosome (BAC) libray of the 4019-20 strain.

The BAC libray comprised 3840 clones, and the insert size ranged from 60 to 228kb, with an average size of 136kb. We generated a total of 5,4,31 expressed sequence tags(ESTs) from the three different stages of F.velutipes, Of these, 3,332 different unigenes were consistent with 73% singlets and 27% contigs, Structural characteristics and phylo-genetic analysis of mitochondrial rDNA cluster were also investigated.
